Serum refers to the colloidal liquid in which fibrinogen is removed from plasma. Every 100ml of human serum contains 6-8g of protein, mainly albumin and globulin. Serum does not coagulate. It has the function of immunity, maintaining acid-base balance and osmotic pressure, and serum protein can also be reserved for use when body protein is insufficient. Human and animal serum is commonly used for various serological tests to help diagnose diseases. Serum containing antibodies can be used for prevention or treatment of diseases.
Main usage
1. When thawing serum, please follow the recommended gradual thawing method (-2 ℃ to 4 ℃ to room temperature). If the temperature changes too much during serum thawing (such as -20 ℃ to 37 ℃), experiments have shown that sediment is very easy to form.
2. When thawing serum, please shake it evenly at any time to ensure uniform temperature and composition, and reduce the occurrence of precipitation.
3. Do not keep the serum at 37 ℃ for too long. If left at 37 ℃ for too long, the serum will become turbid, and many unstable components in the serum will also be damaged, affecting the quality of the serum.
4. The thermal inactivation of serum is very easy to cause an increase in sediment, and if not necessary, this step may not be necessary. [3]
5. If thermal inactivation of serum is necessary, please follow the principle of 56 ℃ for 30 minutes and shake evenly at any time. Excessive temperature, prolonged time, or uneven shaking can cause an increase in sediment.
If flocculent precipitates are found after thawing the serum, the serum can be divided into sterile centrifuge tubes and centrifuged slightly at 400g. The supernatant can then be added to the culture medium and filtered together. However, it is not recommended to remove these flocculent sediments through filtration as it may clog the filtration membrane.
